Cosmetic Peptides: Three Groups, Three Mechanisms
Cosmetic peptides fall into three groups that work on completely different principles. Some interrupt nerve to muscle signalling. Some feed the machinery that builds structural protein. Some act on pigment. Knowing which group a compound belongs to tells you more than any product description will.
Group one: signal blockers
Expression lines come from muscle movement. Repeated contraction of the small muscles of the face folds the skin over them, and over time those folds become permanent.
To make a muscle contract, a nerve releases neurotransmitter into the gap between them. That release depends on a group of proteins called the SNARE complex, which pulls the vesicle containing the neurotransmitter up against the cell membrane so it can fuse and release its contents. No SNARE assembly, no release, no contraction.
Snap-8
Snap-8 is acetyl octapeptide-3, an eight amino acid peptide. Its sequence is modelled on the end of SNAP-25, one of the proteins in the SNARE complex.
The idea is competition. By resembling part of SNAP-25, it competes for a position in the complex, and a complex assembled with an imposter in it works less efficiently. Less efficient assembly means less neurotransmitter released and weaker contraction.
It is the successor to Argireline, acetyl hexapeptide-8, which works the same way with a shorter sequence. Snap-8 is the extended version.
Worth being precise about the comparison to botulinum toxin, since these are constantly described as alternatives to it. They target the same system from different angles. Botulinum toxin is an enzyme that cleaves SNARE proteins outright, which is why its effect is profound and lasts months. A competing peptide interferes with assembly rather than destroying the components. Same pathway, very different magnitude.

Group two: matrix builders
The other reason skin changes with age has nothing to do with muscle. It is the structural protein underneath.
The dermis is built from collagen and elastin, produced by fibroblasts. Production declines with age while breakdown continues, and the result is loss of firmness and elasticity.
GHK-Cu
The most researched compound in this group. GHK is three amino acids bound to a copper ion, and it occurs naturally in plasma at levels that decline with age.
The copper is the functional part. It is the cofactor for lysyl oxidase, the enzyme that cross links collagen and elastin fibres, and cross linking is what converts loose protein strands into tissue with real structural strength. It is also the cofactor for the copper zinc form of superoxide dismutase.

Two things separate this group from the first. Signal blockers act on movement and their effect stops when they do. Matrix builders act on the tissue itself, which is slower and structural rather than functional.
Group three: pigment
The third group acts on melanin, and it runs through the melanocortin system.
MT-1
MT-1 is an analog of alpha-MSH, melanocyte stimulating hormone. It acts on MC1R, the melanocortin receptor found on melanocytes, and activating it drives melanin production.
This is where the peptide families on this site connect in a way worth noticing. Alpha-MSH has produced three separate compounds pulling in three directions:
- MT-1 keeps the pigmentation activity through MC1R.
- PT-141 keeps the central arousal activity through MC4R in the brain.
- KPV is the three residue tail carrying the anti-inflammatory activity, and it leaves pigmentation behind entirely.
One parent hormone, five receptors, three fragments developed for three different purposes. MT-1 is stocked in 10 mg vials.
The other direction
Pigment research runs both ways. Glutathione inhibits tyrosinase, the rate limiting enzyme in melanin synthesis, and shifts production toward the lighter form of melanin. Same system, opposite end.

They do not overlap, which is why they are found combined in blends rather than chosen between.
