Estrogen-related receptors

The estrogen-related receptors, ERRα, ERRβ and ERRγ, are a group of proteins called orphan nuclear receptors. Orphan means no natural hormone has been firmly established as their trigger, which sets them apart from the actual estrogen receptor.

They got their name from a structural resemblance to the estrogen receptor, not from a shared function. What they actually do is regulate energy metabolism. They control the expression of genes involved in mitochondrial function, oxidative metabolism and how cells handle fuel, and they are most active in tissues with high energy demand such as muscle and heart.

SLU-PP-332 is a pan-ERR agonist, meaning it activates all three of them rather than selecting one.

Why that resembles exercise

When you exercise, muscle cells face an energy demand and respond by ramping up their capacity to produce energy. Part of that response runs through the ERR pathway, driving the genes that build and maintain mitochondria and that favour burning fat for fuel.

Activating ERR directly switches on some of that same programme without the mechanical work. That is the logic behind the exercise mimetic label. The research reports increased oxidative metabolism and increased fatty acid use, which are hallmarks of the adaptation exercise produces.
