Amino Acid Blends Explained: Why They Are Not Peptides

Amino acid blends sit in the same catalogs as peptides and get discussed as though they work the same way. They do not, and the difference is structural rather than a matter of degree.

A peptide is a chain with a sequence, and that sequence is what fits a receptor. A free amino acid is a single unit with no sequence to read.

Why the distinction changes what to expect

BPC-157 works because its particular fifteen residue order interacts with specific pathways. Change the order and it stops being BPC-157.

Free amino acids do not work that way. They enter the same pools your diet supplies and get used as raw material: built into proteins, converted into other molecules, or burned. So the question with an amino acid blend is never which receptor it binds. It is whether supply of that particular building block was limiting anything in the first place. The nitric oxide group

Arginine, citrulline and ornithine turn up together constantly, and they are three points on one cycle rather than three separate ideas.

Arginine is the substrate nitric oxide synthase uses to produce nitric oxide, the signal that relaxes the smooth muscle in blood vessel walls and widens them. That is the same molecule PDE5 inhibitors work downstream of, which we get into in how PDE5 inhibitors work.

Taken by mouth, citrulline raises blood arginine more reliably than arginine itself does, because arginine gets heavily metabolized by arginase in the gut wall and liver before it reaches circulation while citrulline bypasses that and is converted to arginine in the kidney. If the goal is arginine in the blood, the indirect route is the more efficient one.

Ornithine sits on the other side of the same cycle, where it takes part in clearing ammonia through the urea cycle. Ammonia accumulates as a byproduct of protein and amino acid breakdown, and the urea cycle is how the body disposes of it.

The structural group

A second cluster shows up in blends aimed at tissue and recovery, and these are building blocks in the literal sense.

  • Proline and its hydroxylated form make up roughly a quarter of collagen by residue count, which is why it appears in anything oriented toward connective tissue or skin.
  • Lysine is an essential amino acid, meaning the body cannot make it, and it is required both for the cross linking step that gives collagen tensile strength and for synthesizing carnitine.
  • Glutamine is the most abundant free amino acid in the body and the preferred fuel of the cells lining the gut and of activated immune cells, which is why it drops during serious illness or injury.
  • Taurine is not built into proteins at all. It works instead on cell hydration, calcium handling and bile salt formation, and sits at high concentration in heart and muscle tissue.

Collagen production also depends on cofactors rather than raw material alone. Vitamin C is required for the hydroxylation step, and copper for the cross linking enzyme, which is the reasoning behind GHK-Cu carrying copper and why approaches to aging skin tend to combine the two ideas.

Carnitine and NAC

Two more components appear often enough to be worth separating out, and neither belongs to the groups above.

Carnitine is the shuttle that carries long chain fatty acids across the inner mitochondrial membrane so they can be oxidized. That transport step is obligatory, which is why carnitine also anchors the lipotropic formulas.

NAC is N-acetylcysteine, and it exists to supply cysteine. Glutathione is a tripeptide of glutamate, cysteine and glycine, and cysteine is the rate limiting one of the three, so raising cysteine availability raises the ceiling on glutathione production. NAC is the stable, absorbable way to deliver it, and it has genuine approved clinical use in paracetamol overdose, where it works by restoring glutathione in the liver. Are amino acids peptides?

They are the units peptides are built from, not peptides themselves. Two amino acids joined make a dipeptide, and a chain of them makes a peptide. A free single amino acid has no peptide bond in it at all.

Why inject them if food contains them?

The usual argument concerns absorption ceilings and first pass metabolism, arginine being the clearest case since much of an oral dose is broken down before it reaches circulation. Does an amino acid blend build muscle?

Muscle protein synthesis responds mainly to the essential amino acids and to leucine in particular, along with the mechanical stimulus of training.
