The point of using a fragment

Full growth hormone raises IGF-1 and affects insulin sensitivity and blood glucose. Those effects come from regions of the molecule that the 176-191 fragment does not contain.

So AOD-9604 does not bind the growth hormone receptor the way intact hGH does, and research has consistently found it does not raise IGF-1 or produce the glucose effects associated with the full hormone. Isolating the fragment isolates one activity and leaves the rest behind. That was the entire design goal.

What it does in fat tissue

The activity studied in adipose tissue runs in two directions. It stimulates lipolysis, the breakdown of stored triglycerides into fatty acids, and it inhibits lipogenesis, the process of building new fat stores.

Work in animal models and in isolated fat cells has connected this to beta-3 adrenergic receptor signaling in adipose tissue, a pathway distinct from the growth hormone receptor route.

AOD-9604 has also been studied outside metabolism entirely, in cartilage and joint research, where the interest is in its effects on chondrocytes rather than on fat.

5-Amino-1MQ: not a peptide

This one is worth stating plainly because the name and the company it keeps suggest otherwise. 5-Amino-1MQ is a small molecule, 5-amino-1-methylquinolinium. It has no amino acid chain and it is not built like a peptide.

It works as an enzyme inhibitor, and the enzyme is NNMT.

What NNMT does

NNMT stands for nicotinamide N-methyltransferase. It takes nicotinamide, a form of vitamin B3, and attaches a methyl group to it, producing 1-methylnicotinamide.

That reaction matters for two reasons. Nicotinamide is a precursor for NAD+, the coenzyme every cell uses in energy metabolism, so methylating it diverts material away from NAD+ production. The methyl group itself comes from SAM, S-adenosyl methionine, which is the cell’s main methyl donor for a long list of other reactions.

NNMT is expressed at high levels in adipose tissue and in the liver. Inhibiting it means less nicotinamide gets consumed and less SAM gets spent, which raises the available pool of both.

What the research has shown

Studies in mice have found that reducing NNMT activity in fat tissue, either by knocking the gene down or by using an inhibitor, produces smaller adipocytes and lower body weight in diet induced obesity models. The effect is tied to increased cellular NAD+ and to changes in how fat cells handle energy.

5-Amino-1MQ is orally available, which separates it from most compounds in this space. Peptides get broken down by digestion, which is why they are almost always injected. A small molecule does not have that problem, and 5-Amino-1MQ is supplied here in 10 mg and 50 mg formats.
