Immune Peptides: Thymosin Alpha-1, KPV and VIP
Three peptides turn up repeatedly in immune research, and they have almost nothing in common structurally. One is 28 amino acids from the thymus. One is three amino acids cut out of a pigment hormone. One is a neuropeptide that also relaxes smooth muscle and helps run your body clock.
What links them is a pattern you see all over this field: take a large molecule that does several jobs, find the part responsible for one of them, and use just that piece.
Thymosin alpha-1
Thymosin alpha-1 is a 28 amino acid peptide cut from a larger precursor called prothymosin alpha. It was isolated from thymus tissue, which is the organ where T cells mature.
The thymus is unusual among organs because it starts shrinking early. It is at its largest around puberty and declines steadily from there, replaced progressively by fat. That process, thymic involution, is one of the clearest structural changes in the aging immune system, and it is why thymic peptides drew attention in the first place.
How it acts
Thymosin alpha-1 works on both halves of the immune system, which is what makes it interesting.
On the innate side, it has been shown to act through toll-like receptors, particularly TLR2 and TLR9. Toll-like receptors are the sensors that recognize patterns associated with pathogens and start the first response. On the adaptive side, research reports effects on T cell maturation and differentiation, along with increased production of signaling molecules including IL-2 and interferon gamma. Effects on dendritic cell function and natural killer cell activity also appear consistently.
It is described as a modulator rather than a stimulant. The research shows it acting on how the immune response is coordinated rather than simply pushing everything harder in one direction.
Thymosin alpha-1 has the most developed regulatory record of the three. Under the name thymalfasin it is approved in a number of countries for hepatitis B and C, and it has been studied as a vaccine adjuvant and in sepsis.

KPV
KPV is three amino acids: lysine, proline, valine. It is the tail end of alpha-MSH, melanocyte stimulating hormone, specifically residues 11 to 13.
Alpha-MSH is best known for pigmentation, but it is also a strong anti-inflammatory molecule. Researchers narrowed down which part carried that second activity, and the answer was this three residue tail. KPV keeps the anti-inflammatory effect and leaves the pigmentation behind.
That should sound familiar. PT-141 came out of the same molecule family by the opposite route, isolating the arousal activity instead. Same parent hormone, two different fragments, two completely different research directions.
How it acts
The mechanism most often reported is inhibition of NF-kB signaling. NF-kB is a transcription factor that sits at the center of the inflammatory response, switching on the genes for inflammatory cytokines. Blocking it acts upstream of a whole set of inflammatory outputs rather than on any single one.
KPV is small enough to enter cells directly, and research describes it acting inside the cell rather than only at surface receptors. In the gut, uptake has been linked to PepT1, a transporter that intestinal cells normally use to absorb small peptides from digested food. A peptide that can hitch a ride on the normal absorption machinery is unusual, and much of the KPV literature covers colitis and inflammatory bowel models for that reason.
Skin inflammation and wound healing form the other main thread, which is why KPV appears in the four way recovery blend alongside BPC-157, TB-500 and GHK-Cu. Available separately in 5 mg and 10 mg vials.
VIP
VIP stands for vasoactive intestinal peptide, a name that captures only a fraction of what it does. It is 28 amino acids long and it was first identified for relaxing smooth muscle and dilating blood vessels in the gut.
It turned out to be far more widespread than that. VIP is found throughout the nervous system, the lungs, the gut and in immune cells themselves, and it acts on two receptors, VPAC1 and VPAC2.
Three separate roles
- Immune regulation. VIP is a strong anti-inflammatory signal. Research describes it shifting the balance of T helper cell responses and supporting regulatory T cells, the population whose job is to hold immune activity in check. Much of the interest is in conditions driven by an immune response that will not switch off.
- Smooth muscle and blood vessels. The original activity. This is the basis for research in pulmonary conditions, where vasodilation in the lungs is the target.
- The body clock. VIP is produced by neurons in the suprachiasmatic nucleus, the brain’s master clock. Its job there is keeping those neurons synchronized with each other. Without VIP signaling the clock cells drift out of phase and the rhythm falls apart.
That last role connects this post to DSIP and sleep. Circadian regulation and immune function are not separate systems, and VIP sits in both.

ARA-290, the same idea again
Worth including because it shows the fragment pattern especially clearly.
Erythropoietin, EPO, is the hormone that drives red blood cell production. It also has a separate protective effect on tissue that has nothing to do with red cells. Those two activities run through different receptors.
ARA-290 is an 11 amino acid peptide taken from the helix B region of EPO. It binds the innate repair receptor, which carries the tissue protective signal, and it does not act on the receptor that drives red cell production. So it carries the protective activity without raising hematocrit. Research has focused on neuropathic pain and on small fiber neuropathy in sarcoidosis. Available here.

Is KPV related to Melanotan?
